Sefazolinin değişen dozlarının in vitro 3T3-Fare fibroblast kültürü üzerine etkisi

Abstract

Amaç: Sefazolin’in değişen dozlarının in vitro fibroblast kültürü üzerine olan etkilerinin araştırılması amaçlandı.

Gereç ve Yöntem: Bu çalışmada, in vitro iyileşme modellemelerinde yaygın olarak kullanılan 3T3 fare embriyonik fibroblast hücre soyu kullanıldı ve sefazolinin 0 μg/ml, 50 μg/ml, 100 μg/ml, 250 μg/ml, 500 μg/ml ve 750 μg/ml olarak belirlenen farklı dozları in-vitro olarak 3T3 fare fibroblast hücreleri üzerine uygulandı. Deneyler 24 ve 48 saat olarak planlandı. Bu sürelerin sonunda her bir grup, CCK-8 hücre proliferasyon, migrasyon ve oksidatif stres testleri ile değerlendirildi.

Bulgular: Gruplara ait IC50 değerleri 24 ve 48. saatler sonunda sırası ile 500- 750 μg/ml doz aralığında olarak bulundu. Buna karşın 50-500 μg/ml doz aralığında uygulanan sefazolinin ise hücre proliferasyonu üzerine olumsuz bir etki göstermediği görüldü. Migrasyon hızının doz ve zaman bağımlı olarak azaldığı tespit edilirken, buna karşın sefazolinin farklı doz aralıklarında uygulandığı deney gruplarında oksidatif stres açısından anlamlı bir fark tespit edilmedi.

Sonuç: Terapötik sınırlarda pre ve postoperatif olarak uygulanan sefazolin in vitro koşullarda 3T3 fare fibroblast hücreleri üzerinde olumsuz bir etki oluşturmamıştır.

Different doses of cefazolin effect on in vitro 3T3-Mouse fibroblast culture

Abstract

Aim: The purpose of this study was to investigate the effects of varying doses of Cefazolin on in vitro fibroblast culture.

Material and Methods: In this study, the 3T3 mouse embryonic fibroblast cell line, which is widely used in in-vitro healing models, was used. Different doses of cefazolin, determined as 0 μg/ml, 50 μg/ml, 100 μg/ml, 250 μg/ml, 500 μg/ml and 750 μg/ml, were applied in-vitro on 3T3 mouse fibroblast cells. Experiments were planned as 24 and 48 hours. At the end of these periods, each group was evaluated with CCK-8 cell proliferation, migration and oxidative stress tests.

Results: The IC50 values of the groups were found to be in the range of 500- 750 μg/ml at the end of the 24th and 48th hours, respectively. On the other hand, it was observed that cefazolin administered in the dose range of 50-500 μg/ml did not have a negative effect on cell proliferation. While it was determined that the migration rate decreased depending on dose and time, no significant difference was found in terms of oxidative stress in the experimental groups in which cefazolin was administered at different dose intervals.

Conclusion: Cefazolin, administered pre- and postoperatively within therapeutic limits, did not cause any adverse effects on 3T3 mouse fibroblast cells in vitro.
