EFFECTS OF VITAMIN E AND CAFFEIC ACID PHENETHYL ESTER (CAPE) ON METHABOLIC ENZYMES OF RATS WITH EXPERIMENTAL LIVER ISCHEMIA-REPERFUSION INJURY

Abstract

In this study, the activities of liver carbohydrate metabolism enzymes, hexokinase (HK), glucose-6-phosphate dehydrogenase (G6PD), 6-phosphogluconate dehydrogenase (6PGD), lactate dehydrogenase (LDH) and malate dehydrogenase (MDH) after experimental ischemia-reperfusion of raf. liver and effects of E vitamin and caffeic acid phenethyl ester (CAPE) on these enzyme activities were investigated. Wistar albino male rats were divided into 4 groups each consisting of 10 rats. Isotonic, CAPE and vitamin E were applied intraperitoneally in the I/R groups. Activities of HK, G6PD and 6PGD were increased and activities of LDH and MDH were decreased in İsotonic+I/R group in comparison with the sham group. İncreased activities of HK, 6PGD and decreased activities of LDH and MDH were observed in vitamin E group compared to isotonic+l/R group. İn CAPE group, activities of HK, G6PD and 6PGD were increased and activity of LDH were decreased significantly when compared to the İsotonic+l/R group. There was no difference between CAPE and sham groups. İncreased activities of HK, G6PD, 6PGD and MDH were detected in CAPE group in comparison with vitamin E group. As a result, we may conclude that CAPE protects glycolysis and pentose phosphate pathway in the injured tissue more than vitamin E and thus decreases injury via increased nucleic acid synthesis as well as supporting energy production by increased activity of citric acid cycle. We conclude that CAPE produces these effects not directly by decreased/increased metabolic enzyme induction, but by indirectly via decreasing tissue injury as shown in our recent studies.
